چکیده

Introduction and Aim: In medicine, the term ‘cancer’ is used to describe a group of disorders defined by uncontrolled cell growth ability invade metastasize other organs. As result body's fight conventional cancer treatments, it essential look for alternative therapeutic avenues. Recent years have seen rise in application reverse genetics techniques including Newcastle disease virus as an oncolytic agent tumor models immunocompetent malignancy. preclinical studies, recombinant NDV (rNDV-GFP) that expresses foreign genes was shown be more effective therapy. The goal this study examine vitro anticancer effects GFP-expressing, genetically modified (rClone3-GFP) strains on JHH5 hepatocellular carcinoma line. Materials Methods: To generate NDV, GFP gene inserted into genome strain (NDVClone30) technology, where now resides region occupied F HN (rNDV-GFP). Using MTT assay, we evaluated rNDV-GFP its potential against cells. Cytopathic were analyzed using fluorescent light microscopy. Results: We observed 96 hours post infection, capable inducing death time-dependent way. Conclusion: Recombinant expressing GFP, demonstrated good results inhibiting growth. Our pave door viral vector treatment liver cancer.

Identification of Optimal Insertion Site in Recombinant Newcastle Disease Virus (rNDV) Vector Expressing Foreign Gene to Enhance Its Anti-Tumor Effect

Recombinant Newcastle disease virus (rNDV) is tumor selective and intrinsically oncolytic, which has been developed as a vector to express exogenous genes to enhance its oncolytic efficacy. Our previous studies found that insertion sites of foreign gene in rNDV vector affected its expression and anti-tumor activities. However, the optimal insertion site for foreign genes remains unknown. In thi...

Tumorcidal Effect of Recombinant Newcastle Disease Virus

We have generated GFP expressing recombinant Newcastle Disease Virus (rNDV-GFP) based on a live vaccine strain which contains a less pathogenic cleavage site of F protein leading to restricted viral growth in vivo. We evaluated the rNDV have an effect of oncolytic activity to B16 melanoma cell. The rNDV-GFP infected efficiency to B16 cells and induced cell death. Apoptosis-related genes, Bax an...

Construction of Recombinant Bacmid DNA Encoding Newcastle Disease Virus (NDV) Fusion Protein Gene

Background and Aims: Newcastle disease virus (NDV) is one of the major pathogen in poultry. Vaccination is intended to control the disease as an effective solution nevertheless this virus is a growing threat to the poultry industry. F gene open reading frame (ORF) from NDV is 1650 bp, encoding a protein of 553 amino acids that can induce protective immunity alone. The F glycoprotein on the surf...

Generation of a Recombinant Akabane Virus Expressing Enhanced Green Fluorescent Protein.

UNLABELLED We generated a recombinant Akabane virus (AKAV) expressing enhanced green fluorescence protein (eGFP-AKAV) by using reverse genetics. We artificially constructed an ambisense AKAV S genome encoding N/NSs on the negative-sense strand, and eGFP on the positive-sense strand with an intergenic region (IGR) derived from the Rift Valley fever virus (RVFV) S genome. The recombinant virus ex...
